RBC Washing

This protocol removes unwanted plasma and plasma proteins, metabolic waste products, microaggregates and platelets from a packed RBC unit using processing/washing solution.

  • Packed RBC protocols end after the Supernatant-Out operation, leaving the ending cell product more concentrated
  • Resuspended RBC protocols end after the Agitate/Wash-In operation, leaving the ending cell product resuspended in processing/washing solution

Deglycerolized Frozen RBC Units

These protocols remove glycerol from thawed RBC units using decreasing concentrations of processing/washing solutions.

The following protocols end after the Supernatant-Out operation, leaving the ending cell product more concentrated:

  • Deglycerolized High-Glycerol Frozen RBCs
  • Deglycerolized Low-Glycerol Frozen RBCs
  • Deglycerolized High-Glycerol Frozen RBCs Low Volume Unit (60 mL to 80 mL)

The Deglycerolized Resuspended High-Glycerol Frozen RBCs protocol ends after the Agitate/Wash-In operation, leaving the ending cell product resuspended in processing/washing solution.

Use with these disposable sets:
  • COBE 2991 Cell Processing Set
  • COBE 2991 Double Coupler Adapter (if required)
Preparation of Leukocyte Concentrates

This protocol can be used to prepare a leukocyte concentrate from a single unit of non-leukoreduced fresh blood.

  • Utilizes differential sedimentation of blood components according to their specific gravities within a gravitational field
  • Accelerates the sedimentation process, allowing the relative isolation of the granulocyte-rich fraction, which may then be expressed into a transfer pack for subsequent use

Use with the COBE 2991 Cell Processing Set

Collection of Young RBCs (Neocytes)
  • Utilizes differential sedimentation of blood components according to their specific gravities within a gravitational field
  • Accelerates the sedimentation process, allowing the relative isolation of the young RBC-rich fraction, which may then be expressed into a transfer pack for subsequent use
  • Allows for the isolation and collection of the younger half of the original unit

Bone Marrow Processing Using a Density Gradient Separation Medium
  • The goal of in vitro manipulation of bone marrow is to reduce the volume and RBC contamination of the bone marrow harvest while minimizing the loss of MNCs, in particular stem and progenitor cells
  • The triple processing set reduces the processing time and the need for multiple single processing sets

  • COBE 2991 Female Luer Connector
Bone Marrow Concentration
  • Prepares a concentrated buffy coat from a large volume of bone marrow; bone marrow concentration is based upon the principle of differential sedimentation of blood components according to their specific gravities within a gravitational field
  • Accelerates the sedimentation process, allowing the relative isolation of the buffy coat, which may then be expressed into a transfer pack for subsequent processing or use
Use with the following disposable sets:
  • COBE 2991 Cell Processing Set
  • COBE 2991 Double Coupler Adapter (if required)
MNC Washing
  • Removes cryopreservative, hemolyzed RBCs or other contaminants from MNC products

Cell Concentration
  • Concentrates expanded cells from large volumes
Use with the following disposable sets:

Large Volume Wash (Shed Blood Salvage)
  • Washes a large volume of salvaged shed blood for autotransfusion or source blood for industrial/commercial use
