ARTICLE: Efficient Manufacturing of Therapeutic Mesenchymal Stromal Cells (MSCs) with the use of the Quantum® Cell Expansion System

This article, authored by Patrick Hanley of Children's National Medical Center (formerly of Baylor College of Medicine) and published in the journal Cytotherapy, evaluates the Quantum system for the cost-effective, safe, and reproducible expansion of large numbers of MSCs from unprocessed bone marrow compared with a flask-based method currently used in clinical trials. The findings of the article include:

  • The Quantum system was able to generate the target cell number of 2.0 × 108 cells in an average of 9 fewer days and in half the number of passages required during flask-based expansion
  • Quantum system-expanded MSCs infused into an ischemic stroke rat model had a significant therapeutic benefit when compared with saline placebo controls, while flask-generated MSCs did not have significant therapeutic benefit
  • It was estimated the Quantum system would involve 133 open procedures versus 54,400 in flasks when manufacturing for a clinical trial
  • MSCs expanded in the Quantum system met the International Society for Cell Therapy (ISCT) standards for expression of CD73, CD90 and CD105 and lack of expression of hematopoietic lineage markers and HLA-DR
